pancreatic cancer, one of the most aggressive tumors, has a dismal prognosis because of the low rates of early identification, fast progression, difficulties following surgery, and the ineffectiveness of current oncologic therapies. there are no imaging techniques or biomarkers that can accurately identify, categorize, or predict the biological behavior of this tumor. exosomes are extracellular vesicles that play a crucial rule in the progression, metastasis, and chemoresistance of pancreatic cancer. they have been verified to be potential biomarkers for pancreatic cancer management. studying the role of exosomes in pancreatic cancer is substantial. exosomes are secreted by most eukaryotic cells and participated in intercellular communication. the components of exosomes, including proteins, dna, mrna, microrna, long non-coding rna, circular rna, etc., play a crucial role in regulating tumor growth, metastasis, and angiogenesis in the process of cancer development, and can be used as a prognostic marker and/or grading basis for tumor patients. hereby, in this concise review, we intend to summarize exosomes components and isolation, exosome secretion, function, importance of exosomes in the progression of pancreatic cancer and exosomal mirnas as possible pancreatic cancer biomarkers. finally, the application potential of exosomes in the treatment of pancreatic cancer, which provides theoretical supports for using exosomes to serve precise tumor treatment in the clinic, will be discussed.
background leukemic cells facilitate the creation of the tumor-favorable microenvironment in the bone marrow niche using their secreted factors. there are not comprehensive details about immunosuppressive properties of chronic myelogenous leukemia-derived exosomes in the bone marrow stromal and immune compartment. we explained here that k562-derived exosomes could affect the gene expression, cytokine secretion, nitric oxide (no) production, and redox potential of human primary cord blood-derived t cells (cb t cells). methods human primary cord blood-derived t cells were treated with k562-derived exosomes. we evaluated the expression variation of some critical genes activated in suppressor t cells. the alterations of some inflammatory and anti-inflammatory cytokines levels were assessed using elisa assay and real-time pcr. finally, no production and intracellular ros level in cb t cells were evaluated using greiss assay and flow cytometry, respectively. results our results showed the over-expression of the genes involved in inhibitory t cells, including nqo1, pd1, and foxp3. in contrast, genes involved in t cell activation such as cd3d and nfatc3 have been reduced significantly. also, the expression of interleukin 10 (il-10) and interleukin 6 (il-6) mrnas were significantly up-regulated in these cells upon exosome treatment. in addition, secretion of the interleukin 10, interleukin 6, and interleukin 17 (il-17) proteins increased in t cells exposed to k562-derived exosomes. finally, k562-derived exosomes induce significant changes in the no production and intracellular ros levels in cb t cells. conclusions these results demonstrate that k562-derived exosomes stimulate the immunosuppressive properties in cb-derived t cells by inducing anti-inflammatory cytokines such as il-10, reducting ros levels, and arising of no synthesis in these cells. moreover, considering the elevation of foxp3, il-6, and il-17 levels in these cells, exosomes secreted by cml cells may induce the fates of t cells toward tumor favorable t cells instead of conventional activated t cells.

background multiple sclerosis (ms) is a chronic condition that primarily manifests as demyelination of neuronal axons in the central nervous system, due to the loss or attack of oligodendroglia cells that form myelin. stem cell therapy has shown promising results for the treatment of ms due to its capability to halt the immune attack, stop apoptosis and axonal degeneration, and differentiate into oligodendrocytes. stem cell-derived exosomes (exosomes) have shown great capabilities for neuronal diseases as they have growth factors, complex sets of mirna, enzymes, proteins, major peptides, lipids, and macromolecules with anti-inflammatory, angiogenesis, and neurogenesis activities. methods this study aimed to compare the healing properties of stem cells, against exosomes for the treatment of an experimentally induced ms dog model. dog models of ms received either a single treatment of stem cells or a single treatment of exosomes intrathecally and the treatment process was evaluated clinically, radiologically, histopathologically, and electron microscopy and cerebrospinal fluid analysis. results showed marked amelioration of the clinical signs in both treated groups compared to the control one, magnetic resonance scans showed the resolution of the hyperintense lesions at the end of the study period, the histopathology and electron microscopy showed marked healing properties and remyelination in treated groups with superiority of the stem cells compared to exosomes. conclusions although stem cell results were superior to exosomes therapy; exosomes have proven to be effective and safe important actors in myelin regeneration, and their use in diseases like ms helps to stimulate remyelination.

alzheimer's disease (ad) is characterized by the accumulation of β-amyloid peptide (aβ) in the brain because of an imbalance between aβ production and clearance. neprilysin (nep) is the most important aβ-degrading enzyme in the brain. thus, researchers have explored virus-mediated nep gene delivery. however, such strategies may entail unexpected risks and thus exploration of a new possibility for nep delivery is also required. here, we show that human adipose tissue-derived mesenchymal stem cells (adscs) secrete exosomes carrying enzymatically active nep. the nep-specific activity level of 1 μg protein from adsc-derived exosomes was equivalent to that of ~ 0.3 ng of recombinant human nep. of note, adsc-derived exosomes were transferred into n2a cells and were suggested to decrease both secreted and intracellular aβ levels in the n2a cells. importantly, these characteristics were more pronounced in adscs than bone marrow-derived mesenchymal stem cells, suggesting the therapeutic relevance of adsc-derived exosomes for ad.
